What is Tofajak-In 5mg (TOFACITINIB)?
JAK inhibitor for rheumatoid arthritis, psoriatic arthritis, ulcerative colitis, and polyarticular juvenile idiopathic arthritis. One of the first approved oral biologics for RA.
How it Works
Inhibits JAK1 and JAK3, blocking signaling of pro-inflammatory cytokines including IL-2, IL-4, IL-7, IL-15, and IL-21 — reduces joint inflammation at the molecular level.

Common Side Effects
Serious infections (bacterial, viral, fungal), lymphoma risk, MACE (major adverse cardiovascular events), venous thromboembolism, elevated lipids, anemia, neutropenia.
Safety Warnings
⚠️ Black box warnings: serious infections, malignancy, MACE, and thrombosis. TB screening mandatory. Do not combine with biologics. Live vaccines contraindicated. Age 50+ with CV risk factors: consider alternatives.
