Compound type: Neuropeptide — OX2R-Preferring Agonist CAS Number: 205120-59-8
What is Orexin B (Hypocretin-2)?
Orexin-B (Hypocretin-2) is a 28-amino acid linear (no disulfide bonds) neuropeptide co-produced with Orexin-A in lateral hypothalamic neurons. Unlike Orexin-A (equipotent at OX1R/OX2R), Orexin-B has ~10-fold preference for OX2R over OX1R, making it the preferred tool for OX2R-selective research. OX2R is particularly important for histaminergic arousal (tuberomammillary nucleus), circadian output, and metabolic regulation. The receptor subtype selectivity between Orexin-A and Orexin-B enables dissection of OX1R vs. OX2R contributions to orexin biology.
Research Applications
OX2R-Selective Research
OX2R preferring (10:1 vs OX1R) — isolates OX2R contribution in orexin biology
Histaminergic Arousal
Tuberomammillary nucleus OX2R for histamine-mediated wakefulness
Circadian Output
SCN OX2R modulation of circadian timing and output
Receptor Comparison
Parallel Orexin-A vs Orexin-B studies to define OX1R vs OX2R pharmacology
Sleep Disorder Models
OX2R-mediated sleep-wake switching in narcolepsy and insomnia models
What is Orexin-B and how does it differ from Orexin-A?
Orexin-B (Hypocretin-2) is a 28-amino acid linear neuropeptide that preferentially activates OX2R (~10-fold over OX1R). Unlike Orexin-A (equipotent at both subtypes), Orexin-B enables OX2R-selective research — particularly for histaminergic arousal, circadian output, and OX2R-specific pharmacological studies.
How is Orexin-B reconstituted?
Add 1–2mL bacteriostatic water to the lyophilized vial. Orexin-B is linear (no disulfide bonds) and dissolves readily to a clear colourless solution. Swirl gently. Store at 2–8°C for up to 21 days.
