What is MOTS-c?
MOTS-c (Mitochondrial Open reading frame of the Twelve S rRNA-c) is a 16-amino acid peptide encoded by the mitochondrial genome — the first peptide discovered to be encoded by mitochondrial (not nuclear) DNA. Functions as a mitokine: secreted from mitochondria, travels to the nucleus to regulate metabolic stress response genes.
How Does MOTS-c Work?
MOTS-c activates AMPK via inhibition of the folate cycle and purine synthesis pathway. AMPK activation drives: fatty acid oxidation, mitochondrial biogenesis (PGC-1α), glucose uptake, and mTOR inhibition. Also directly regulates nuclear genes via Nrf2 and FOXO3 pathways.
