Mechano Growth Factor (MGF) is a splice variant of IGF-1 generated by alternative splicing to include the E peptide domain from exon 5 (Ec in humans). Unlike systemic IGF-1-Ea, MGF is locally expressed in response to mechanical loading — acting as an autocrine/paracrine signal for satellite cell (muscle stem cell) activation and proliferation. The E-domain peptide is the active component, binding non-IGF-1R targets to drive satellite cell pool expansion independently of systemic IGF-1 pathways. MGF expression is highest immediately post-exercise and declines rapidly — motivating PEG-MGF development for extended half-life research.

How is MGF reconstituted?
