What is IGF-1 LR3?
IGF-1 LR3 (Long R3 Insulin-like Growth Factor-1) is a recombinant analog of IGF-1 with two modifications: substitution of Arg for Glu at position 3 (reduced IGFBP binding) and addition of a 13-amino acid N-terminal extension. These changes reduce IGFBP-3 binding by ~500-fold, extending half-life from ~12 minutes to ~20–30 hours.
How Does IGF-1 LR3 Work?
IGF-1R binding → IRS-1/IRS-2 phosphorylation → PI3K/Akt/mTOR → protein synthesis and anti-apoptotic signalling. MAPK/ERK pathway → cell proliferation. Reduced IGFBP binding means LR3 remains biologically active in tissues where native IGF-1 would be sequestered.
