What is FOXO4-DRI?
FOXO4-DRI is a D-amino acid retro-inverso peptide that disrupts the FOXO4-p53 protein-protein interaction. Senescent cells (characterised by p21/p53 expression) use this interaction to avoid apoptosis. FOXO4-DRI breaks the interaction, triggering apoptosis specifically in senescent cells while sparing healthy cells.
How Does FOXO4-DRI Work?
Senescent cells overexpress FOXO4, which sequesters p53 in the nucleus and prevents pro-apoptotic PUMA expression. FOXO4-DRI is a competitive inhibitor of FOXO4-p53 binding → releases nuclear p53 → PUMA upregulation → mitochondrial apoptosis. Normal cells lack the FOXO4 overexpression required for this pathway.
